Hemorrhagic cysts of the anterior pituitary were first developed in intact, female rats by the prolonged administration of a pituitary preparation containing gonadotrophins (Selye, Collip and Thompson, '33). Subsequently, enlargement of the pituitaries of female rats was obtained with the administration of an extract of pregnancy urine or estrin (Wolfe, Phelps and Cleveland, '33 ; Wolfe, '35a and b), and adenomatous tumors of the anterior lobe were found to develop in rats and mice following several months of estrogen treatment (Cramer and Horning, '36; McEuen, Selye and Collip, '36; Zondek, '36). A large body of experimental evidence on the effects of short and long term estrogen administration on the cytology and physiology of the anterior pituitary has since been published, and has been summarized by several authors (Zondek, '41; Wolfe, '49; Gardner, '48 and ' 5 3 ) . The present report is an attempt to relate the observations on estrogen-induced pituitary tumorigenesis obtained in our laboratory to the extensive work of others, and to present a working hypothesis which may be of use in studies on the etiolo-gy of pituitary tumor formation.
