The diagnosis of congenital infections is hampered by the poor development of the child's immune system and transplacental passage of maternal antibody. The correspondents describe anin situhybridization method for the diagnosis of HIV infection in the newborn. Mononuclear cells were isolated from a mother and baby by density centrifugation and cultured with phytohaemagglutinin and interleukin-2 for 2 weeks.35S-labelled HIV-specific cDNA was used to probe the lymphocytes. After one week 3.4% of maternal cells, and 3.8% of the infant cells were positive. At the end of 2 weeks <1% of maternal cells were positive whereas 3.9% of the baby's lymphocytes were still positive. This method is presented as a means of providing an early diagnosis of congenital HIV infection.newline˜S.H. Gillespie
