About 55% of the lysosomal enzyme N-acetyl-.beta.-D-glucosaminidase (NAGase) in newly collected mastitic milk was present within the cellular compartments, the centrifugate or the top cream layer. In milk from healthy quarters this percentage was even higher, 94%. The NAGase was released from the cellular compartments upon storage, a cycle of freezing and thawing, by stimulating cells with endotoxin or opsonized zymosan. Maximum release of activity was obtained by including 2% Triton X-100 in the milk samples. NAGase analysis requires standardization of the sampling and handling of milk samples. Monitoring the release of NAGase into the skimmed milk fraction is suggested as a method for analyzing cellular activation.
