Male adults of Drosophila, which do not normally produce yolk polypeptides are stimulated to do so by 20-hydroxy-ecdysone. The response to a single dose of hormone is a short burst of yolk-polypeptide synthesis by the fat body. The magnitude of the response is dose dependent. A 2nd treatment with the hormone leads to a similar response. Long-term feeding of males does not lead to permanent activation of the yolk-polypeptide genes. It was not possible to stimulate male fat bodies to synthesize yolk polypeptides with 20-hydroxy-ecdysone in vitro. 20-Hydroxy-ecdysone, possibly in the presence of another hormone or a carrier protein, induces transcription of the yolk-polypeptide genes of the male fat body and its presence is required to keep the genes active. In females mature fat bodies can produce yolk polypeptides in vitro without the requirement for added hormone. Male and female fat bodies have different hormonal requirements for the maintenance of expression of the genes coding for the yolk polypeptides.
