A study was carried out to determine whetherP. cariniitrophic forms use the mitogen-activated protein kinase (MAPK) pathway to regulate the transition into cysts and whetherP. cariniihave pheromone receptors as part of the MAPK machinery participating in life cycle regulation. Pheromone receptor expression onP. cariniiorganisms was detected by FACS.P. cariniiMAPK PCM gene expression under basal conditions was at low levels compared to theP. cariniiactin housekeeping gene. TheP. cariniiMAPK PCM mRNA expression was increased four times in trophic forms compared to cysts. The PCM kinase from the trophic forms had a significantly increased activity compared to that from the cysts. A cDNA sequence with significant homology to the fungal pheromone receptors, in particular to theS. cerevisiae STE3pheromone receptor, was identified. This gene is reported in Genbank under the accession number AY007236. A subpopulation ofP. cariniitrophic forms expresses this receptor. These data show that theP. cariniiMAPK PCM is differentially regulated in theP. cariniilife cycle, exhibiting greater mRNA expression in trophic forms compared to cysts, and possessing greater kinase activity in trophic forms compared to cysts.
