This paper is from the Medical Research Division of the Upjohn Company. Clinimycin, a derivative of lincomycin, is 7(S)-chloro-7-deoxylincomycin hydrochloride hydrate, and has been shown to possess higherin vitroantibacterial activity than lincomycin, especially against staphylococci. This paper reports only a pharmacological study in young normal adult males. Briefly, it shows that Clinimycin is very rapidly absorbed, the peak level being reached in 45 minutes in fasting subjects: its half-life is 2.38 hours, that of lincomycin being reported to be 5.2 hours. Administration at 4 different times in relation to a meal showed that food only delayed absorption and did not impair it. It was equally well absorbed from capsules and tablets. Only about 10% or less of the dose was recovered from the urine: the fate of the remainder is unexplained. The findings are reported in great detail, and are complicated by the fact that one known and " possibly one or more" unknown derivatives of lincomycin are formed in the body: a second series of assays was done with extracts of serum or urine containing Clinimycin only.
