An estrogen binding protein present in the renal cytosols of untreated normal, castrated and estrogen-treated hamsters has a sedimentation value of 4S by sucrose density gradient analysis when compared to ovalbumin and bovine serum albumin. The amount of binding was enhanced with prolonged estrogen treatment. The binding site exhibits specificity for estrogenic hormone (e.g., diethylstilbesterol, estrone, estriol, estradiol-17α) since 3H-estradiol binding is decreased markedly in the presence of these unlabeled estrogens but not with dihydrotestosterone, aldosterone, cortisol, progesterone and metabolites of progesterone. The nature of this estrogen binding macromolecule was further ascertained by its precipitation with protaminesulfate. Although this specific macromolecular binding by estrogens in the kidney is much less than in the hamster uterus, it is considerably greater than that in the plasma, lung or liver.
