To the Editor: In their interesting article, Dr. Duvoisin et all make the point that pergolide may be more effective than bromocriptine in treating patients with advanced Parkinson's disease, because pergolide was much less dependent on presynaptic dopaminergic elements for its effect. The authors based this latter conclusion on the observation that after pretreatment with alphamethyl-paratyrosine, rats treated with pergolide continued to have more circlingactivity than those treated with bromocriptine. Although these results were impressive, I question whether they reflect any underlying physiologic property rather than merely a pharmacologic manipulation. The authors state that the dosage of 0.25 mg/kg pergolide is a t least four times more potent than 1.0 mg/kg bromocriptine. It may be the case that if the authors had tested equipotent doses of these agents in the alphamethyl-paratyrosine experiment, the responses of the animals in the two groups would have been similar.
