A method is described for transplanting a nucleus from a blastula cell into a previously enucleated frogs egg. Eggs of Rana pipiens. pricked but not enucleated, showed an abortive type of cleavage, whereas eggs pricked and enucleated showed no cleavage. 99% of 638 enucleations were successful. Of 498 embryos obtained from normally inseminated previously enucleated eggs, all but one were haploids, indicating the success of the operation. When enucleated pipiens eggs were injected with a diploid pipiens blastula cell nucleus more than half cleaved and gave rise to partial and complete blastulae. 74% of the complete blastulae gastrulated normally, forming complete embryos, of which about half were normal and the remainder slightly abnormal. Cytological evidence for removal of the egg nucleus was obtained by fixing 22 eggs at various stages. Since it was known that diploid or haploid embryos produced by crosses between R. catesbeiana and R. pipiens died at early stages of development, the transplantation technique was tested by placing catesbeiana nuclei in pipiens enucleated eggs. The donor blastulae were produced by inseminating pipens eggs with catesbeiana sperm and letting part of the eggs develop as diploid hybrids, whereas in others the egg nucleus was removed, giving androgenetic haploid hybrids. Transplantations involving both the diploid and haploid blastula cells resulted in death at the same time as the controls. Cytological studies of hybrid control and exptl. blastulae showed similar abnormalities, except for 3 cases in the latter where the nuclei were in an abnormal interphase condition characterized by diffuse Feulgen staining. Use of the technique for studies of nuclear differentiation and function is discussed.
