The action of trypsin used to separate myocardial cells before their plating is specified. Different cellular types in the seeding deposit are identified. In culture the myocardial cells of new-born rats appear to undergo 2 successive stages following the cell dissociation of the organ. Up to the 24th h of culture, a stage of disorganization of cellular structures is observed, characterized by the appearance of disorder in the internal organization of the myoblasts and by the formation of new structures. This stage of disorganization is followed by a stage of reorganization and differentiation, during which the progressive and concomitant maturation of certain organelles is accomplished (evolution of fasciae adhaerentes, the setting of the trails of myofilaments and sarcomerogenesis). The cellular differentiation increases from 48 h of culture, a stage during which the whole culture rhythmically contracts.
