By meristem treatments with 0.4% colchicine and seed treatments with 02, 0.4, and 0.8% colchicine mature plants with a double number of chromosomes were obtained of the following Nicotiana species and hybrids: N. tabacum, N. rustica, N. glauca, F1 (N. tabacuto X. N. glauca), F1 (N. tabacum X N. sylvestris), F1 (N. glutinosa X N. glauca), and some N. rustica X N. tabacum segregants. The autotetraploids of N. tabacum, N. rustica and N. glauca were characterized by smaller plant habit, smaller and thicker leaves with larger stomata and increased pollen sterility compared to the diploids. The polyploids of N. tabacum and N. rustica had larger seed than the diploids. Allotetraploids of the sterile F1''s (N. tabacum X N. glauca) and (N. tabacum XN. sylvestris) were fertile, had wider and thicker leaves with larger stomata and were similar in growth habit to the F1''s. Allotetraploids of the dwarf hybrid, N. glutinosa X N. glauca, showed a marked increase in plant vigor, leaf size and pollen fertility over the F1. Hyperpolyploids of N. rustica X N. tabacum segregants had smaller plant habit, smaller and thicker leaves with larger stomata and increased pollen fertility compared to un-doubled sister plants.
