MATERIALS AND METHODSCats (1.5-2.0 kg) were anesthetized by intraperitoneal injection of 50 mg/kg of sodium pentobarbital. Right ventricular papillary muscles were rapidly removed and placed in a IO-ml muscle bath containing Krebs-Ringer solution aerated with 95% 02 and 5% CO2 at 29 C. The muscles were stimulated at a rate of lZ/min, with a pulse duration of 5-7 msec at a voltage 10% above threshold. The muscles were fixed below in a spring-loaded clip, which was a rigid extension of a tension transducer, and were attached above by a wire clip to the end of an isotonic lever system (9). Initial muscle length was set by a small preload and kept constant by a micrometer stop. After at least 1 hr of stabilization within the bath, both passive and active length-tension curves of the muscles were obtained, and three preloads selected from the rising portion of the active curve. GE force-velocity and SE load-extension curves were obtained at these preloads at 29 C. Temperature was then lowered to 24 C and the SE and force-velocity curves obtained for the same preloads at that temperature, and again at 29, 34, and 37 C. In addition, four muscles were studied at 22 C, and one studied with temperature first raised to 37 C and then lowered to 24 C. Lengthtension curves were determined at 24 C and 34 C in three muscles.
