Figure S2. A) DNA polyplex stability in serum determined by agarose gel shift assay at N/P ratio of 16 and different FBS serum concentrations. DNA polyplex formation (in 20 mM HEPES pH 7.4) and incubation time (30 min) was performed as for the in vivo experiments. Serum was added to the polyplexes after particle formation and the samples were incubated for 15 min at room temperature. Polyplexes with 80 were completely stable, with 78 some very moderate destabilization (smear) was observed. Naked DNA (“DNA”) shows some degradation (disappearance of supercoiled form) and some retardation at highest FBS concentration
