Transport of protein between all compartments of the secretory pathway of eukaryotic cells is governed by the sequential formation (budding) and fusion of carrier vesicles. These vesicles control both the constitutive and regulated flow of protein between the endoplasmic reticulum (ER), the Golgi and the cell surface. The biochemical and molecular basis for vesicle budding and targeting, and the mechanisms fundamental to vesicle-membrane fusion have become the subject of intense scrutiny with the advent of model in vitro systems to study these partial reactions for each stage of the secretory pathway, and the development of a genetic system for dissecting the pathway in yeast. As the basic properties of vesicular transport have been recently reviewed [1, 2], the present discussion will focus only on new developments in our understanding of the biochemical and molecular basis for transport of protein between the early compartments (ER and Go@) of the secretory pathway.
