In a wide variety of cells, Ca2+-mobilizing receptors on the surface membrane stimulated by agonists activate phospholipase C. The activation of phospholipase C appears to be mediated by guanine nucleotide-dependent regulatory protein(s), and results in the hydrolysis of phosphatidylinositol 4,5-bisphosphate. This hydrolysis leads to the production of inositol 1,4,5-trisphosphate [(l,4,5)IP3] and diacylglycerol (DG). (l,4,5)IP3 stimulates the Ca“ release from an intracellular pool and DG activates calcium-phospholipid-dependent protein kinase. Therefore these molecules are considered second messengers. The purpose of this review is to summarize some current ideas on the relationship between phosphoinositide metabolism and Ca“ mobilization in parotid salivary gland.
