BackgroundThe release of platelet-activating factor (PAF) as a fluid-phase mediator from IgE-sensitized rabbit basophils was first re ported nearly 10 years ago [1, 2], Since that time, many studies have been conducted to characterize PAF and have implicated its release in mediating acute allergic reac tions [cf „3, 4], PAF is a potent stimulator of washed rabbit platelets inducing a cal cium-independent shape change and a cal cium-dependent platelet aggregation and secretion of granular constituents [5], PAF-induced platelet aggregation and se cretion previously were shown to be inde pendent of intrinsic ADP release [6] and independent of the production of prosta glandins or their intermediates [7, 8]; sub sequently, however, PAF stimulation of platelets was shown to initiate platelet prostaglandin production and thrombox ane Bi release [8, 9] and to induce the re lease of ADP which enhanced platelet aggregation but not secretion [10],
