The breakdown of chlorophyll in suspension cultures of tobacco cells (Nicotiana tabacum var. "Samsun") placed in the dark without shaking at different temperatures was investigated by in-vivo spectroscopy and chromatography (HPLC) of the extracts. With increasing time and/ or temperatures of incubation of the cells the Soret band shifts to shorter wavelengths indicating pheophytinization in the cells, but afterwards this shift is reversed to longer wavelengths. The pattern of the elution chromatogram of the extracts from senescing cells changes only in the relative amount of the different pigments: chlorophyll a, pheophytin a, pheophorbide a, a more polar pheophorbide a called "X", sometimes chlorophyllide a and two other minor peaks. "X" is assumed to be 13-hydroxypheophorbide a as concluded from the spectroscopic (UV-Vis, MS), chromatographic (TLC, HPLC) and chemical (conversion to the methylester) similarities with an authentic sample.
