It has been suggested that the synaptic ribbon is functionally equivalent to the dense projections in conventional synapses (Gray and Pease, 1971) and that it functions to convey the synaptic vesicles in an orderly fashion to the presynaptic membrane by way of filamentous-like striations connecting the ribbon with the vesicles (Bunt, 1971). This report examines the filamentous connections of photoreceptor synaptic ribbons in the chick retina using the ethanolic phosphotungstic acid (E-PTA) technique of Bloom and Aghajanian (1966) and describes further ultrastructural features of these ribbon filaments which may be related to their proposed function.Chick retinas from 19-day old embryos and 2-day old hatchlings were fixed by ocular perfusion with 2% glutaraldehyde (Ladd, 70%), 2% paraformaldehyde, 0.5% acrolein, 0.5% dimethylsulfoxide in 0.08 M phosphate buffer (Skoff and Hamburger, 1974). The lower temporal quadrant was dissected out and stored in the same fixative for 18 hr at 4 C and then hand dissected into smaller pieces, dehydrated and stained 1 hr with agitation in 1% PTA in absolute ethanol, containing three drops of 95% ethanol per 25 ml staining solution (Bloom and Aghajanian, 1966). The tissue was then dehydrated in absolute ethanol, rinsed in propylene oxide, and embedded in Epon-Araldite. Thin sections were cut on a diamond knife and examined without further staining in a Phillips EM 301.
