Incubation of 131l-labelled serum proteins with guinea pig tissue slices in vitro led to the formation of firmly bound protein-tissue complexes. Desorption of significant amounts of adsorbed protein required the use of strong urea solutions. Saturation of the available tissue surface with protein required the use of a high concentration of protein in the medium (there were a large number of available sites for protein adsorption). When antibody was used as an adsorbate, the bound antibody could react with antigen indicating that the bound protein remained in a superficial position. Iodoacetamide inhibited the adsorption process suggesting that tissue thiol groups are required.
