The important anatomic position of the cornea, its characteristic immune response to foreign tissues and antigens and the com plex physiologic activity of its several layers in the maintenance of corneal transparen cy have rendered this tissue a valuable test object for investigations in many different fields. Many of these studies require the use of a single anatomic layer of the cornea. Sheets of pure corneal endothelium have been prepared and cultured by a number of investigators. 1" 6 Since the corneal stroma occupies a considerable thickness of this tis sue, it can be separated without difficulty by the excision or peeling off of the underlying Descemet's membrane and overlying corneal epithelium. The preparation of sheets of pure corneal epithelium, on the other hand, poses a somewhat more difficult problem. While tissue-culture techniques have been used to grow pure corneal epithelium, 3· 7'8 it has thus far been impossible to separate pure sheets of epithelium from the intact cornea.
