Many secretagogues, hormones and neuro-transmitters produce their effects by a mechanism that involves a rapid increase in intracellular calcium levels. The calcium is released from intracellular vesicular stores by the action of inositol trisphosphate (IP3). IP3 is a cytosolic second messenger cleaved from phosphatidyl-inositol-4,5-diphosphate present in the cell membrane by the agonist-activated enzyme phospholipase C also resident in the membrane. IP3 is short lived, being quickly dephosphorylated within the cell to di- and mono-derivatives before being reincorporated into the cell membrane.
