Isolation of Marine Birnavirus from Sea Squirts Halocynthia roretzi

Mass mortality of the culturedHalocynthia roretzihas been occurred in south coastal regions of Korea since 1995. We isolated a virus (UR1) from diseasedH. roretziin May of 1999 and conducted virus characterization, serological and molecular biological investigations. UR1 was isolated from digestive gland and pharyngeal slit in chinook salmon embryo (CHSE-214) and rainbow trout gonad (RTG-2) cell line. The virus particles were unenveloped, hexagonal in appearance and had an average diameter of 63 nm. The RNA of UR1 consisted of 2 segments as a characteristic of birnaviridae. The UR1 was serologically similar to MABV strains other than IPNV. Reverse transcription-polymerase chain reaction (RT-PCR) and sequencing of the 470bp of VP5, 770bp of internal VP2 region, and 643bp of VP3 region on genome segment A were carried out. The homology of UR1 with MABV Y-6 and NCI was over 97.5 % in all the regions mentioned above. It was concluded that the isolate from aH. roretziis a member of MABV. It is the first report of a virus isolation from ascidians.
