Real time functional MRI (rtfMRI) neurofeedback has been suggested as a scientific tool for improving individual cognitive function. Using this method, we recently found that working memory performance could be improved by self-regulating the brain activity in the dorsal lateral prefrontal cortex (DLPFC). However, the neural effect of the rtfMRI neurofeedback training is not very clear. Increasing evidence from healthy subjects and clinical patients has indicated that the cortico-subcortico-cerebellar circuit is closely related to working memory. In this study, we concentrated on the effect of rtfMRI training on this cortico-subcortico-cerebellar circuit. The results showed that the functional connectivity in this circuit was significantly improved during the training. Specifically, the thalamus, an integration region in the circuit, showed the most improved connectivity with other regions. Moreover, the improvement in connection strengths exhibited a positive correlation with the promotion of working memory updating task performance. The findings in this study advance our understanding of brain mechanism for optimizing working memory function by neurofeedback training. The increases in circuit connectivity by rtfMRI training give some implications for repairing the impaired connection in this circuit in patients with working memory deficits in the future.
