The gastrin-releasing peptide (GRP) is a 27 amino acid neuropeptide and growth factor that is homologous to the amphibian tetradecapeptide bombesin.'*2 The amidated carboxyl-terminal heptapeptide, which is identical in the two peptides, represents the biologically active region' that is necessary and sufficient for binding to high-affinity cell surface receptors.' GRP is found in mammalian brain,5 where it influences body temperature, satiety, and many other central homeostatic functions! GRP is also widely represented in peripheral nerves surrounding the gut,' where it stimulates the release of a number of gastroenteropancreatic hormones' and modulates gastrointestinal motility.' In addition, GRP is found in fetal neuroendocrine cells of the peribronchial epithelium," as well as in neuroendocrine cell tumors from the human lung and thyroid, namely small cell lung cancer (SCLC)," pulmonary carcinoid tumor,'* and medullary carcinoma of the thyroid." Under certain circumstances, GRP stimulates the growth of some cultured SCLC cell lines,lcI6 normal human bronchial epithelial cells," and the Swiss 3T3 mouse embryo fibroblast cell line.'' In Swiss 3T3 cells, after the GRP mitogenic signal is initiated by binding of GRP to high-affinity cell surface receptors, it is subsequently transduced through a complex series of second messengers, including an elevation of intracellular pH and changes in intracellular free calcium, phosphatidylinositol metabolism, and protein kinase C activity." These events are followed by transient transcriptional induction of the c-myc and c-fos genes," increased DNA synthesis, and cell division. In some cultured human SCLC cell lines, concentrations of GRP as low as I nM elicit a rapid and transient increase in intracellular free calcium2' and phosphatidylinositol turnover:2 which can be blocked by a monoclonal antibody ( 2 A l l ) directed against the carboxyl terminus of the peptide.
