RESULTSEfficient engraftment of NPCs occurred in all 1.5 injected mice. The injections did not cause significant morbidity or abnormal neurological function. In the first day after injection, the NPCs were localized in the subventricular zone and fluid-filled spaces of the brain. At 7 days, labeled cells were found widely dispersed throughout the CNS, suggesting they had migrated from the initial sites of engraftment. The NPC did not appear to disrupt the architecture of the brain. The cells mingled with host neurons and glia. In fact, the only distinction between NPCs and host neurons and glia was the presence of cytoplasmic and perinuclear electron-dense X-gal precipitates. Labeled (37.2 cells were identified in 89% of the coronal brain slices examined from mice 7 to 21 days after injection (Fig 1).
