A light-heavy fractionation of pig liver microsomes is described. The light microsomes are rich in cytochrome b5 and evidence is presented that these are the microsomes appearing as smooth-surfaced vesicles in the electron microscope; also that these smooth-surfaced microsomes are biochemically and morphologically distinct from rough-surfaced microsomes. Descriptions are given of effects of solubilizing agents and low pH on microsomes, of the binding of hemoglobin, of possible seasonal effects, and of microsomal pigments, including one which when reduced complexes with CO to show an absorption peak at 450 m[mu]. A microsomal substance (probably a breakdown product) which rapidly oxidizes cytochrome b5 is described, and its possible significance is discussed.
