Controlled Interfacial accumulation of tricyclic antidepressant drugs on carbon surfaces provides the basis for sensitive and selective voltammetric measurements of these compounds. Cyclic voltammetry Is used to aid In the Interpretation of re-sults. Adsorption and extractlon/adsorption are shown to be the mechanisms of preconcentration at the glassy carbon and carbon paste electrodes, respectively. The relative extent of accumulation depends on the structure of the antidepressant drug. The differential pulse response Is evaluated with respect to electrolyte, electrode material, preconcentration period, concentration dependence, detection limit, reproducibility, possible interferences, and other variables. After 4 min of preconcentration, detection limits of 1.4 X 10~ 8 M trimipr-amine, 1.5 X 10~ 8 M imipramine, and 1.7 X 10~ 8 M desipr-amine are obtained. Transfer of the electrode, with the ac-cumulated drug, to a blank solution Is used to eliminate in-terferences due to solution-phase electroactive species. Applicability to direct measurements In physiological fluids Is Illustrated.The tricyclic antidepressants desipramine, imipramine, and trimipramine are the most widely used drugs for the treatment of depression. These drugs are theorized to enhance nora-drenergic activity throughthe blockade of norepinephrine reuptake in peripheral and central noradrenergic neurons. Their efficacy in alleviating depression or side effects depends upon their concentration inphysiological fluids. Because of the low levels of tricyclic antidepressants in these fluids, highly sensitive techniques are required for their quantification.
