The effect of triiodothyronine (T3) on cerebroside deposition and on the incorporation of [35S]sulphate into lipids was studied in the rat brain between 9 and 26 days after birth. Cerebroside depositions was accelerated to a small extent and only before 17 days of age. The incorporation of35S into brain lipids was maximal at 17 days, but the peak was less pronounced in the T3-treated than in the control animals. At the age the specific radioactivity of the separated [35S]sulphatides was reduced by treatment with T3, but the reduction was no longer apparent when the data were expressed in terms of the concentration of acid-soluble35S. There was also an anomaly even in the control between the rates of cerebroside deposition and of the incorporation of [35S]sulphate into lipids during the period from 17 to 26 days. It was concluded that the content of [35S]sulphatides is not a valid measure of myelin deposition during development, unless the kinetics of incorporation of35S into the brain are also taken into account.The investigation also included a study of the age distribution of35S in the fractions containing mucopolysaccharides and acid-soluble constituents in the brains of both the control and T3-treated animals. The maximum content of35S occurred at 17 days in the fraction containing mucopolysaccharides but at 13 days in the acid-soluble fraction. The result showed that factors determining the availability of precursor sulphate must be considered in assessing relative rates of35S incorporation.
