The report by P orter (1) that no free-a-amino group is detectable in ovalbumin by the dinitrofluorobenzene method of S anger (2) has led to numerous efforts to characterize the N-terminal structure of ovalbumin. Using both S anger's method and E dman's phenylisothiocyanate method (3), T itani* of the author's laboratory has carefully repeated the N-terminal analysis of ovalbumin and has likewise found no evidence for the presence of an unsubstituted N-terminal amino acid.
