A purification method is described for gene 20 product which is a quantitatively minor, but essential, protein of the bacteriophage T4 head. During purification and in response to different solvent conditions the protein behaves as if it were hydrophobic. Its structure was studied by conventional and scanning transmission electron microscopy and sedimentation analysis. Gene 20 product exists as a dodecameric structure with 12-fold rotational symmetry and a MW of .apprx. 740,000 as measured by 2 independent methods (electron scattering and sedimentation equilibrium). It is composed of a hollow cylinder, 14 nm long and 7 nm diameter, and 12 radial protrusions, each measuring .apprx. 7 nm .times. 5 nm. Double rings and microcrystals of rings were observed. The microcrystals measure .apprx. 1 .mu.m and have a body-centered tetragonal type of lattice. The structure of the gene 20 product oligomer, in particular its symmetry, has consequences for the assembly of the T4 head, whose shell has 5-fold symmetry.
