In this determination, the oxidation of glucose in the pres-ence of glucose oxidase at pH 7 provides a source for H202 which, in turn, reacts with Fe (CN) 63~ in the presence of lu-minol (3-aminophthalhydrazide) to produce chemilumines-cence (CL) proportional to the initial glucose concentration. CL emission is observed only in basic solution, and an am-monia buffer of high capacity Is used to rapidly adjust the pH of the solution from levels of high enzyme activity to lev-els of detectable CL emission. The integrated CL signal ob-tained after five minutes of enzyme activity was linear with glucose concentration in 10-µ samples over the entire range of ordinary clinical interest (2-200 mg/dl). Because the method relies on an emissive process rather than ab-sorbance, it may find applicability in the rapid determination of small quantities of glucose in biological tissues.
