H~+ Involving Signal Transduction in Regulation of Vicia faba L. Stomatal Movement by Jasmonic Acid

Using fluorescent probe BCECF-AM and Fluo-3 monitored the changes of pHi and [Ca2+]cyt in guard cells of Vicia faba under confocal microscopy. Whether H+ was involved in signal transduction in JA regulating stomatal movement was tested. The results showed that (–)JA induced stomatal closure at concentrations of 0.1~100 μmol·L-1 was mediated intracellular alkalization in guard cells. In contrast, (±)JA induced stomatal open without pHi changes. Pharmacological experiment proved that Na3VO4 reduced the effectiveness of (–)JA- induced stomatal closure, while fusicoccin had no effect on (±)JA-induced stomatal opening. Compared changes of [Ca2+]cyt induced by (–)JA and (±)JA: (–)JA increased [Ca2+]cyt rapidly, while (±)JA induced [Ca2+]cyt increas- ing and decreasing slowly. These results demonstrated that (–)JA and (±)JA had different roles and signal pathways in regulation of stomatal movement.
