An anesthetic dose of urethane was found to be very effective in blocking spontaneous ovulation in the rat when administered just prior to or during the proestrous critical period. Its principal action appears to be blockade of LH—RH release from the brain. The drug does not act at the pituitary to alter significantly the gland's responsiveness to exogenous LH—RH. If administered ip urethane also appears to block spontaneous ovulation by a peripheral action: approximately 5 times as much exogenous LH is needed to ovulate urethane—blocked rats as pentobarbital— bioeked rats. Even when LH levels in the blood are elevated well above values necessary for full ovulation under pentobarbital, ip urethane can block ovulation. An iv injection of urethane does not block ovulation under these conditions, and it appears to lack the peripheral blocking effect.
