0’2 2 ml of 1-9% PCA is added to 0 1 1 ml platelet-rich plasma or serum and the mixture is incubated at 370C for 20 min. After centrifugation at 3000 rpm for 15 min, 0-ml of 0 2 2 mol/I sodium hydroxide is added to 0-1 ml of the supernatant. Endotoxin is assayed with a commercially available reagent consisting of lyophilised amoebocyte lysate from Tachypleus tridentatus and the synthetic chromogenic substrate Boc-Leu-Gly-Arg-p-nitroanilide (’Pyrodick’; Seikagaku Kogyo Ltd, Tokyo).
