The Ly-5 alloantigenic system discovered by Komuro and co-workers (1975) has recently been discussed in detail (Scheid and Triglia 1979, Michaelson et al. 1979). This antigen has been detected on a variety of hematopoietic cells including T and B lymphocytes and lymphomas, plasmacytomas, peritoneal macrophages, monocytic tumor lines, myeloid cells [including neutrophils (Potter et al. 1979), eosinophils and basophils (MAS Moore, personal communication)], prothymocytes, cytotoxic effector cells (Woody et al. 1977), and" natural killer" cells (Kasai et al. 1979). It is absent, however, from erythrocytes, erythroblasts, proerythroblasts, sperm, a neuroblastoma, a mammary carcinoma, and a teratocarcinoma. The Ly-5 phenotype of less differentiated erythrocytic cells has not yet been elucidated. The present study centered around the ontogeny of Ly-5 antigen expression. Using a PA-SRBC rosetting assay, the percentages of Ly-5 § cells in neonatal and adult thymus and spleen were found to be almost identical (unpublished results). Therefore, the following investigation was conducted to pinpoint when during embryogenesis Ly-5 could first be detected in the murine hematopoietic system. Ly-5 expression was quantitated by serum absorption assays and PA-SRBC assays (techniques described by Scheid and Triglia 1979) on fetal tissues obtained from congenic hybrid embryos at various stages of gestation using the presence of a vaginal plug as day 0. The results reported here show that Ly-5 is detectable as early as day 8 of gestation in yolk sac and day 12 in fetal liver, the earliest ages studied.
