A small sample (usually 5 [mu]l) is diluted with dimethylformamide, frozen at liquid N2 temperature, and then reacted with a mixture of pyridine, hexamethyldisilazane, and trimethylchlorosilane. The proportions of the anomeric forms of the fully trimethylsilylated carbohydrate are determined by gas chromatography. The method gives values for the mutarotation coefficients of glucose, galactose, arabinose, and xylose which are identical to those determined by polarimetry. Application of the method to assay of the enzyme, mutarotase, is described.
