Apolipoprotein (apo) E was first discovered more than 25 years ago [1, 2] and plays a central role in the transport of lipids in the plasma and in the maintenance of cholesterol homeostasis [3]. ApoE also plays important roles in normal metabolism and functioning of the central nervous system and in neuropathology [3]. This 299 amino acid, 34 kDa protein occurs in three common isoforms, which are products of three alleles at the same gene locus [4]. The products of these alleles, apoE2, apoE3, and apoE4, differ at two positions, amino acids 112 and 158: apoE3 has cysteine at position 112 and arginine at position 158, whereas apoE2 has cysteine at both positions and apoE4 has arginine at both positions [3, 5]. The amino acid substitutions in the apoE isoforms have an impact upon plasma cholesterol levels, their lipoprotein receptor interactions, and their associations with Alzheimer’s disease [3, 5, 6]. The apoE2 allele appears to be associated with a decreased risk and the apoE4 allele is associated with an increased risk [6] of developing Alzheimer’s disease [7], as compared to apoE3.
