METHODSMale Sprague-Dawley rats, weighing 120-140 g, were housed in rooms in which the artificial lights were switched on at 6 AM and switched off at 6 PM. Fed animals are defined as rats which had free access to water and food (rat laboratory chow) and which were killed between 8 and 9 AM. Starved animals are defined as rats which, having been maintained on their normal diets, were deprived of food from 8 AM of the 1st day of the experiment. In certain experiments rats were force fed glucose by stomach tube while under light ether anesthesia. Animals were killed by exsanguination through the abdominal aorta while under ether anesthesia.The medium used for the assay of heart and adipose tissue LPL activity was essentially that described by Borensztajn et al.(2), except that an activated artificial triglyceride emulsion (Intralipid 10%) replaced chylomicrons as the substrate. The substrate was activated by incubating 1 part Intralipid with 2 parts rat’s serum and 3 parts 0.9% saline for 30 min at 37 C. Adipose tissue LPL activity was measured in acetone-ether dried-powder preparations as described by Wing and Robinson(17), with the modification that the fresh tissue was homogenized in 1 ml rat serum. The heart enzyme activity was determined in fresh tissue homogenized in 0.025 N NHa-NH&l buffer at pH 8.1 (2). The incubation procedures, extraction, and titration of free fatty acids released in the assay medium by the adipose and heart tissue preparations have been described (2). The enzyme activities are expressed in units= t standard deviation, 1 unit representing 1 pmole free fatty acids released per hour incubation per gram wet weight tissue.
