In a recent article, Nakagawa et al.[1] developed a novel filtration method to isolate sperm from forensic samples for sperm-DNA genotyping using a 1: 1: 40 solution of 1% acid fuchsin, 1% methylene blue, and 1 M HCl which they called Baecchi’s staining solution. In 1912, Baecchi, an Italian forensic scientist, wrote an extensive work in German [2] regarding the use of methyl blue as a blue dye while denying the use of methylene blue, and this work is regarded as the authoritative article for this method in more recent studies [3-6]. While methylene blue and methyl blue sound similar, they are chemically distinct substances (Fig. 1-A, 1-B)[7]. Thus, the solution described in Nakagawa et al.[1] is not Baecchi’s staining solution and should have been described more precisely (eg, modified Baecchi’s staining solution with methyl blue replaced by methylene blue).To further clarify the naming and composition of Baecchi’s staining solution, it should be stated that Baecchi initially reported the use of methylene blue in 1909 [8] and that other reports and textbooks in Japan have attributed Baecchi’s name to a double staining solution (s) or method (s) using methylene blue in a 1: 1: 40 solution of 1% acid fuchsin, 1% methylene blue, and 1% HCl (about 0.28 M with specific gravity of 1.02)[9, 10], which has a composition of 0.024% acid fuchsin, 0.024% methylene blue and 0.95% HCl [11, 12], and also to the use of acid fuchsin and methylene blue without a specified recipe or composition [13, 14].
