The properties of a temperature-sensitive mutant ofBacillus subtilisin which DNA synthesis is specifically and reversibly inhibited above 40° are described. When incubated at 45° there was no detectable breakdown of DNA in the mutant and the synthesis of deoxynucleoside triphosphates was normal. The temperature-sensitive element affects DNA synthesisin vivobut notin vitroand (unlike an initiator) it appears to be needed for DNA synthesis throughout the replication cycle.
