Nerve growth factor (NG F~ is required for nornml developmon and maintenance of function of the peripheral sympathetic nervolts system. Administration of mui-NGF antibody to neonates leads to irreversible destruction of their sympathetic ganglia (immunosympathectomy). But axonal transport of NGF is not limited to adrenergic neurons. This report presents a summary of recent findings showing that endogenom NGF is essential for both development mtd fimction of peptidergic semor)'neurons. Specific NGF action on dorsal root ganglia neurons appears to be modulated by glucocorticoids. This phenomenon is atudogous to the characteristic action of glucocorticoids on NG F induced effects in sympathetic ganglia, in adrenal medulhtrv and in rat pheochromocytoma cells.Nerve growth factor (NGF) was originally discovered in the course of investigations on the growth-promoting effects of certain mouse fibrosarcomas. Mouse sarcoma tissue grafted into the body wall of chick embryos produced an increase in the size of sensory and sympathetic ganglia. Levi Montalcini and Hamburger (1951) showed that this tumor was densely innervated by sensory and sympathetic nerve fibers and that the effects were due to a humoral factor produced by the sarcoma tissue. A specific bioassay was developed which measured NGF in a semiquantitative fashion by determining" its ability to produce outgrowth of nerve fibers from explanted chick embryo sensory ganglia. This bioassay made it possible to determine the presence of NGF in a variety of tissues and body fluids including the submandibular gland of the adult male mouse, snake venoms, prostate glands of the guinea pig, rabbit, bull and sheep, and in seminal plasma of the bull and other species. The submandibular gland of adult male mouse was found to contain very high concentrations of NGF and it still represents the major source for isolating this growth factor. The protein extracted from mouse submandibular glands is a multicomponent protein known as 7S NGF.
