Opiates acutely inhibit neuronal firing and hyperpolarize the membranes of most responsive cells studied (for reviews see North, 1979; Henderson, 1983). These effects are probably direct, the result of altered membrane channel conductance or pump activity. Because they cause long-term changes in organisms manifested as tolerance and dependence, one might suppose that opiates effect long-term biochemical changes in cells as well. What changes these are we have little idea at present. Calcium, cyclic AMP (cAMP), and cyclic GMP (cGMP) may act as second messengers to transmit opiate information into cells and initiate or inhibit cellular processes.
