Helene Sage,* Federico M. Farin, Gary E. Striker, and Aron B. Fisher abstract: Primary cultures of rat alveolar type II epithelial cells (granular pneumocytes) producedseveral components of the pulmonaryextracellular matrix. Fractionation by ion-exchange chromatography of radiolabeled protein secreted into the culture medium resulted in the partial purification of two of these components: fibronectin and type IV procollagen. Identification of these proteins by sodium dodecyl sulfate-polyacrylamide gel electrophoresis was confirmed by radioimmune-precipitation studies with affinity-purified antibodies. Thrombospondin, a platelet a-granule protein that was recently shown to be secreted by endothelial and other mesenchymally derived cells and may be involved in platelet aggregation, was, in addition, purified by elution from diethylaminoethylcelluloseIVIore than 40 different cell types have been described in adult human lung [for a review, see Kuhn (1976)]. This cellular heterogeneity reflects not only the structural diversity of the lung architecture in toto but also the metabolic and functional properties of this organ as well. In recentyears, an increased understanding of the primary gas-exchanging
