Female BDF1mice were exposed for 3 weeks to lead in the drinking water at concentrations ranging from 0 to 1000 ppm lead. In vitro mitogen studies demonstrated that lead produced a dose-dependent enhancement of the activity for the T-cell mitogens concanavalin A, phytohemagglutinin, and staphylococcal enterotoxin A. Lead did not alter the activity of the B-cell mitogen, Escherichia coli lipopolysaccharide. A significant synergistic interaction occurred between lead and concanavalin A or phytohemagglutinin, suggesting that lead may be affecting a specific T-cell subpopulation. In vivo mitogen studies with staphylococcal enterotoxin A demonstrated that lead has no significant effect on the mitogen response. Comparisons and extrapolations between the in vivo and in vitro mitogen responses with similar lead exposures are not meaningful.
