Fresh tumor tissue, obtained in the operating room, was processed by a variation of the procedure described by Lubs and Clark.1 Using reagents from a TC chromosome culture kit, the tissue was minced in Hanks solution with added colchicine, and the finely divided bits of tissue were stirred mechanically to release single cells. The supernatant fluid containing the cell suspension was decanted and then handled according to an adaptation of the Moorhead technique2 for leukocyte cultures. Six to eight slides were prepared in each case. They were systematically scanned and every cell which appeared suitable was photographed for chromosome counting and karyotyping, if possible.
