MATERIALS AND METHODSFunctional efficacy of the microencapsulated islets was determined using five totally pancreatectomized diabetic dogs as recipients without immunosuppression. Defined quantity of microencapsulated islets from outbred mongrel donors was grafted through the catheter into omental tissue of the pancreatectomized recipient at 3-to 7-day intervals until a total dose of islets was achieved that restored fasting normoglycemia without exogenous insulin injection. Fasting blood glucose was monitored daily, using chemstrip tapes and precisely determined by a Beckman glucose analyzer type 2 every 7 days. Intravenous glucose tolerance tests (0.5 g glucose/kg body weight) were performed preoperatively and at 1-month postimplantation. K values (decline of glucose level,%/min) were calculated from plasma glucose levels at 5, 10, 30, and 60 minutes after injection of glucose.
