The sensitivity to acetyl-choline (ACh) of chronically denervated mammalian skeletal muscle was studied by iontophoretic application of the agent to single muscle fibers while membrane potentials were simultaneously recorded. In an innervated muscle, ACh produces membrane depolarization only when applied to the end-plate region of the muscle fiber and the area sensitive to ACh occupies only a very limited part of the muscle membrane. In a denervated muscle the entire membrane becomes sensitive to ACh and wherever it is applied on the membrane it produces a depolarization of similar magnitude and time course as when applied to the end-plate area of an innervated muscle. The effect of ACh on membrane permeability to Na and K is the same in denervated muscle as at the normal end-plate. The use of anticholinesterases in denervated muscle does not potentiate the effects of ACh. Although the whole membrane in a denervated muscle is sensitive to ACh, the muscle fibers remain electrically excitable.
