Both haloperidol and arecoline induced a cataleptic state in rats and a marked synergistic effect was observed upon combination of threshold doses of these two drugs. Atropine sulphate, but not atropine methylnitrate, blocked the cataleptic effects of the two drugs alone and in combination.Bilateral lesions of the caudate-putamen potentiated the cataleptic effect of arecoline whilst markedly reducing the cataleptic effect of haloperidol. Only the potentiated effect of arecoline was observed upon administration of a combination of the two drugs to animals with bilateral caudate-putamen lesions. Unilateral lesions of the caudate-putamen had no significant effect upon arecoline catalepsy but partially reduced the cataleptic effects of haloperidol and the combination.The possible role of the extrapyramidal system in these effects is discussed.
