Although the etiology and pathogenesis of multiple sclerosis (MS) are unknown, it has long been believed that aberrant immune function may play a role.’ Within recent years the concept that suppressor T lymphocytes may be reduced in the disease has received considerable emphasis. During acute attacks reduced Con A-induced suppressor cell activity2 and reduced T gamma cells’ have been described. Impaired production of interferon has been In addition, some investigators have observed abnormalities of the natural killer cells: but this area is somewhat controversial.6 Subsequent to the development of monoclonal antibodies which recognize different subsets of T lymphocytes’ it was reported that the subset defined by the OKT5 antiserum was reduced in patients during acute episodes of MS.’ This was believed to be consistent with the notion of deficiency of suppressor cells in MS because the population of Con A-induced T cells which suppress the allogeneic mixed lymphocyte reaction’ was shown to be OKTS positive. Reduced OKT5 cells were also reported by Bach et al.’*; subsequently, the OKT8 antiserum was introduced as a marker for the same set of lymphocytes defined by OKT5. With this reagent, Hauser et al.” provided evidence that variation in lymphocyte subsets correlated with disease activity. However, recently others” have failed to find changes in lymphocyte phenotypes during acute episodes of MS. For the past three years lymphocyte phenotypes have been examined in patients being investigated by the Neuroimmunology Branch. In order to interpret the findings in patients, considerable effort has been devoted to the study of lymphocytes in normal individuals. The purpose of this report is to describe our findings to date in both normals and MS patients.
