The first part’of this two-part review provided an introduction to hematopoietic cell culture, and exam-ined some of the important growth factors and physicochemical parameters that affect culture performance. Emerson has recently reviewed several aspects of hematopoietic culture and its therapeutic applications”. In this second part, we provide an introduction to the field of hematopoietic stem cell transplantation, and examine possible applications of ex vivo expansion of hematopoietic cells. Potential applications include ‘graft engineering’(to produce more rapid short-term engraftment after transplantation), tumor purging, gene therapy, generation of dendritic cells for immunotherapy and the production of mature blood cells for transfusion (Fig. 1).
